How to discover lukewarm (LKW) leucine concentration
« on: January 17, 2017, 09:00:57 AM »
Hi!!
In the lab I'm working there is a vial containing a lukewarm (LKW) leucine solution that was given from another lab, but I don't have any information about the concentration or anything! I asked around and no one had this written. Is that a way to find out the concentration?  ???
I don't know if it helps but the lab has a scintillation counter. I'll need this to measure bacterial production by the 3H-leucine incorporation into proteins.

I assume that leucine is the only amino acid present.  If you need the concentration of leucine, then you might be able to obtain it colorimetrically, with a quantitative ninhydrin assay.  The other possibility that comes to mind is to analyze for nitrogen in some way.  However, I don't know how the presence of tritium changes the practicality of either experiment.
