Title: I want to know principle of DNA crosslinking
Post by: koreanraichu on March 23, 2017, 05:05:15 AM
When oligonucleotide spotted on amin coated glass, that is ionic interaction.
Then we have to bake glass(80 degree, 80min) or UV crosslinking. I want to principle of these reaction, but never answered that.
But I know just 'baking or UV crosslinking is recommend', and 'at dry oven'. I baked dry sample and humidified sample, dry sample shown fluorescence, one the other hand, humidified sample never shown fluorescence.

If I thought right, -NH2+(terminal of aminosilane glass) and PO4-(DNA) reacted with heat and dry environment, some molecules come out such as water?
