Title: Questions concerning biochemical techniques (Quite Challenging!!)

I am trying some of my past exam questions, I have a lot of questions.

I don't know how to separate mRNAs from Total RNAs.

and there is one more question that I could not even understand its idea.

Also, what is Amplification Refractory Mutation System? In its use in PCR system, how do it works?

I don't know how to separate mRNAs from Total RNAs.
Most mRNAs have a polyA tail and therefore bind quite happily to a oligo-dT column (affinity chromatography), while the rest of the RNAs can be found in the flow-through.

In a question from my pastpapers, it is mentioned that there is a possibilities to detect, by PCR, different kinds of mutations, it stress the absence of need to do any post - PCR modification or analysis.

Is it really possible?

I don't know many about PCR techniques and protocols (just the regular stuff), but I think there are some PCR techniques that detect point mutations like those (I mean yes, that is possible). Maybe implying fluorescent markers... ???. Can someone bring some light to the subject??
