Title: UV-VIS measurement

I need to measure UV assay of a drug substance. I measure standard material 3 times and chcek the calibration linear with a control solution. Then I create average of standards. Should I calculate only from 3 standards or calculate with control as well?

You will have to define what you mean by control better.  Do you mean a separate standard prepared as a check standard, or do you mean bracketing standards, the same prep as beginning standards, that are run after the samples?
