Title: TLC Question
Post by: JPinkman on September 13, 2019, 09:22:43 AM
Hello everybody, first time poster here.

I briefly just have a quick question for all.

Does anyone have any experience using the USP method for analyzing Lactose Anhydrous via TLC? I seem to have trouble preparing the solvent system properly leading my plate to have an unstable solvent front.

The preparation is Ethylene Dichloride:Glacial Acetic Acid:Methanol:Water [10:50:30:20]. I have been told if you observe a cloudy solution then there is something off with your ratios. So I have tried pipeting the solutions accurately as well as preparing the acetic acid, methanol and water separately and slowly adding it to the ethylene dichloride with stirring.

I feel like I am missing something right in front of me so if anyone has any advice it would be greatly appreciated.

thanks!
