Title: Sample preparation for Chromium quantification in algae cell

I´m a limnologist, and I´m working with a Selenastrum (algae) in a bioassy under Chromium contamination. I`m asking for help: Wich is the best digestion program for decomposing algae cells, and wich is the best analytical method to quantify the concentration of the chromium accumulated in the Selenastrum cells.

US EPA's SW-846 has a method for sediment that will adapt well. Grinf the sample in a tissue grinder first.

Use inductively coupled argon plasma atomic emission spectroscopy for the determinative step. If you use AA you'll have some matrix troubles. Also, the absorption vs conc function is curved if the flame isn't carefully adjusted.
