Title: organic chemistry of Drugs synthesis?
Post by: Cocain on December 03, 2007, 06:02:47 AM
Can someone help me please? I don't know the way to synthesize promethazine (an antihistamine drugs) and where to find some information about this drugs!

Hi,

For what reason do you want to synthesise this drug? If you are a student or work for industry you wouldn;t be asking this question..

More info and we might help you.

Prometazine is a relatively simple derivative of phenothiazine.
Just find a textbook on drug synthesis in library, eg
The Organic Chemistry of Drug Synthesis. by Daniel Lednicer.

I read the book "The Organic Chemistry of Drug Synthesis. by Daniel Lednicer." but it provides me only some general information about Promethazine. My professor asks me to know at least 3 ways to synthesis P., i've just know only one. I've found on Internet for a long time but no more useful information. Do you know some useful internet links? Thanks!

Phenothiazine can be obtained in very different reactions. In some of them you can add side chain before cyclization.
Try this way.

The side chain is branched. you can try a different approach to its synthesis

Yeah, thank mod very much! I'll try that way. But I think my methods are only used in lab. However. my professor wants the processes in details which are applied in industrial chemistry. I think it's very difficult for a student like me!!! I tried to look up in some patent websites but there is only promethazine HCl. Can someone help me please?

Industrial processes were patented. Prometazine.HCl is a real drug used. Prometazine itself is a base, hence this is only neutralization reaction: amine + HCl. Dont worry about this.
