Title: 5 Kb and 4.95 kb dsDNA strand exchange???
Post by: JoshA on June 03, 2008, 06:57:00 PM
Hi everyone,
I have been looking in the literature but I have not been able to find anything on this subject. I have PCR'ed a 5 kb DNA and a 4.95 kb DNA. The 5 kb and 4.95 kb are identical except that the 5 kb is just 50 bp's longer.
If I put both of these DNA's (5 kb and 4.95 kb) in a test tube and heat them to 100 C for and 1-2 hours, do you think I could get some of the strands from the 4.95 and 5 to swap? This would give me some dsDNA with a 50 bp overhang, which is what I want.
If I can not do this, is there any other way I can go about it? Thanks-Josh
