Title: DNA gel electrophoresis
Post by: lufthoved on August 11, 2005, 06:35:09 AM
Hi  everyone...

Im working with DNA and PCR and wanted to test my amplicons. Therefore I made a gel.

Actually it turned out very good but I was wondering how to improve my result.

1. There a large white shadow behinds my band (Im using EtBr and dye). How can I reduce this (or better yet remove it)?

2. Should I run longer time, so the bands will end up post the shadow?

Pardon my english (I come from Denmark)...

Best wishes

You think the band will be better then, and the shadow smaller?

1.You can try to reduce the amount of ethBr.
2.Sometimes is it enough to clean the photo-table.
3.Running the gel longer makes no sense in my opinion.

best wishes kira

Hi, all. Thank you for your answers.

I was wondering if it would be better could mix EthBr with the samples and apply to the wells instead of mixing in the gel?
