Title: 1D and 2D Chromatography

Anybody knows the difference between 1-dimensional chromatography and 2-dimensional chromatography? What do you think is the advantage of using 1-D or 2-D chromatography? Please advise me. Thank you. :D

In some cases, the chemistry within a given column can be insufficient to separate some analytes. It is possible to direct a series of unresolved peaks onto a second column with different chemical properties. The sample is spotted at one corner of a square plate,developed, air-dried, then rotated by 90° and redeveloped in a second solvent.

So you can seperate certain compounds by using two solvents, that otherwise would not have seperated.

2-d chromatography is also very helpful in telling whether you have a compound that reacts on the plate. If you spot your compound in one corner of a square plate (as SirRoderick suggests) and run it up in one solvent system, then turn it 90o and develop it again in the same solvent system, all of your spots should be along a diagonal. If you get extra spots off the diagonal, then one of your components is reacting on the plate.
