Title: THP deprotection time span
Post by: zzzisoo1 on May 11, 2013, 07:42:23 PM
Hi guys.
I am doing THP deprotection with THP protected alcohol and ppts in ethanol at 70°C for 24 hours.
I was just wondering if it is a big problem to leave it a bit longer than 24 hours, maybe 36 hours.

Thanks =]

No this should not make a difference.

It depends on how sensitive your product is to those reaction conditions.

In my experiance (which includes THP ethers) some of the deprotection methods are very generic so if you take a bit of time to do a TLC then you might find that it is done in an hour or two, or after 36 hours you may find that it is not complete so you need to keep at it.
