Two questions on doing SPE

When doing SPE, what happens in the case I forget to load a portion of my sample in a cartridge and it "dries" under a mild vacuum for a minute or two? Is it safe to just continue with the procedure, or it might impact the outcome of the analysis?

I'm not doing a precise quantification of an analyte at the moment, but the question might just apply in general.

Is it normal to see some "bubbles" in the cartridge when the sample is passing through (pic related, outer portions of the sorbent, right cartridge)? This happens when a cartridge does not dry out, i.e. it is not related to the situation in the question above. Thanks!
