Enzyme Catalyzes Reaction Mechanism

(1/1)

Nejji:
Hi

I have been trying to find out the mechanism of an enzyme catalyzed reaction. It has two substrates and is inhibited by one of the substrates at higher concentrations. By looking at primary plots it seems to have Compulsory-Order Ternary Complex Mechanism. It is enough or do I need product inhibition studied as well to confirm it?
Thanks!!

Babcock_Hall:
My understanding is that the plots of rate versus concentration are not usually considered to be definitive in establishing the kinetic mechanism.  Product inhibition and inhibition by analogs are two additional tools.  Articles in Methods in Enzymology might be of help.

The book Enzyme Kinetics and Mechanism by PF Cook and WW Cleland might be a good resource.
