Please help me!
« on: December 05, 2006, 05:03:45 AM »
Hi everybody :)
I am new here. I want to ask you. I hope you can help me.  Why do we use p-chloromercuribenzoate-agarose affinity column( if I want to purify an enzyme)?
Thank you!

What is unclear: afinity chromatography or p-chloromercuribenzoate? The latter easily bind to the matrix (yielding HCl) and the -COOH group on the other end of the molecule is a common one when it comes to immobilise a protein.

For the former, check this out: http://users.rcn.com/jkimball.ma.ultranet/BiologyPages/A/AffinityChrom.html

Short but clear, I believe.
