Solvent extraction
« on: March 15, 2007, 07:53:35 PM »
Good day,

I usually do hydrocarbon extraction from my culture using an equal amount of dichloromethane  as an extraction solvent. Previously, i had been using 10 ml of culture. So, extraction using 10 ml of dichloromethane is very manageable. Now, I started to set-up a 30 ml culture. Would it be Ok to do extraction using 10 ml of dichloromethane? In solvent extraction, is it a protocol to do extraction using the same amount of solvent?

Could you help me on this matters...

Many thanks

Usual procedure is to use smaller volume of the solvent, but to do extraction in three steps. This way extraction goes much closer to completion.
