using distilled water blank solution?

for an enzyme assay, i never tried yet since i just followed the experiment instructions but what will be the difference if i blank the spectrophotometer only with distilled water instead of blanking it with the reagents+distilled water except the enzyme? Maybe only with distilled water is a faster way to blank it?

The reagents will read as more dilute than they were in solution before being put in the spectrophtometer.

Dear hpl912;

It seems that you haven’t realised, that ignoring the reagents or other ingredients may easily be a big source of important systematic errors.

For an explanation you may read:   "Blank Experiment (#16’463)”  I wrote a little time ago.
