DCM woes!! F-moc solid phase peptide synthesis
« on: June 01, 2013, 05:17:51 PM »
Hello,
I'm doing solid phase F-moc peptide synthesis. As soon as I perform the resin activation with DCM for 2 hrs, the resin comes up in the DCM layer and does not stay on the stage of peptide column. Also this happens after the addition of first amino acid.
What could be the reason for the resin to come up in DCM? Please explain in detail.
P.S. I have washed the peptide column with 0.1 N HCl, acetone and water (and dried for over a day. All the syringes, measuring cylinders, and pipettes are also acetone washed and oven-dried!!
